Heat Shock Proteins and the Regulation of Apoptosis

چکیده
Since the elucidation of their functions in protein folding and translocation, heat shock protein chaperones have been a target of research in all spheres of biomedicine. Within the last five years, research efforts have intensified, following the discovery of raised levels of heat shock protein (Hsp) expression in the brains of patients suffering from many neurodegenerative disorders, including Alzheimer’s, Parkinson’s and Huntington Disease and cerebral ischaemia. Expression of Hsps in the brains of patients is thought to form part of a general protective stress response. The stress in question, however, varies, depending on the particular disease. For example, accumulation of α-synuclein aggregates in Parkinson’s Disease causes stress to the protein folding machinery of the cells, with consequent up-regulation of stress proteins including Hsps. When markers indicative of the occurrence of apoptosis were also found in degenerating brain tissue, the question of how heat shock proteins might impact on apoptotic neural cells was raised. However, their particular function under diseased conditions remains unclear. This chapter highlights the involvement of Hsps in the regulation of neural apoptosis, from the original reports of Hsp expression during neurological disorders, to evidence of their neuroprotective properties and their potential as therapeutic molecules.

Epigenetic alterations, including DNA acetylation, hypermethylation and hypomethylation, and the associated transcriptional changes of the affected genes are central to the evolution and progression of various human cancers, including pancreatic cancer. Cancer-associated epigenetic alterations are attractive therapeutic targets because such epigenetic alterations, unlike genetic changes, are po...
متن کاملDendrosomal Nanocurcumin Induces Changes in the Expression Levels of Heat Shock Proteins in the AGS Cell Line: Cellular Tolerance or Smart Apoptosis Induction
Objectives:The expression levels of heat shock proteins (HSPs) are elevated in many cancers, and this overexpression is often associated with both a poor survival and a therapeutic outcome. Curcumin is an anti-cancer agent that also induces a heat shock response. HSPs confer resistance to curcumin-induced apoptosis in cancerous cells. The aim of the current study was to analyze variations in th...
متن کاملModulation of H2O2- Induced Neurite Outgrowth Impairment and Apoptosis in PC12 Cells by a 1,2,4-Triazine Derivative
Introduction: Increased oxidative stress is widely accepted to be a factor in the development and progression of Alzheimer’s disease. Triazine derivatives possess a wide range of pharmacological activities including anti-oxidative and anti-in.ammatory actions. In this study, we aimed to investigate the possible protective effect of 3-thioethyl-5,6-dimethoxyphenyl-1,2,4-triazine (TEDMT) on H2O2-...
متن کاملHydrogen sulfide treatment protects against renal ischemia-reperfusion injury via induction of heat shock proteins in rats
Objective(s): Hydrogen sulfide (H2S) attenuates ischemia-reperfusion injury (IRI) in different organs. However, its mechanism of action in renal IRI remains unclear. The present study investigated the hypothesis that H2S attenuates renal IRI via the induction of heat shock proteins (HSPs).Materials and Methods: Adult Wistar rats were subjected to unilateral renal ischemia for 45 min followed by...
متن کاملThe Relationship between the Expression of Small Heat-Shock Proteins and Suppression of OCT4B1
Background: OCT4B1, a variant of OCT4, is expressed in both cancer cells and tissues. This variant has a main role in the regulation of both apoptotic and stress (heat-shock proteins) pathways. The aim of this study was to investigate the effects of OCT4B1 silencing on the expression of small heat-shock proteins (sHSPs)in three human tumor cell lines. Methods:...
متن کاملFeeding Artemia larvae with yeast heat shock proteins 82 (HSPs82) to enhance the resistance against abiotic stresses (hyperosmotic and high temperatures)
Feeding farmed Artemia with yeast heat shock proteins is a novel way to protect them from stress conditions during the culture. In this study, the effect of feeding with stressed new identified Saccharomyces cerevisiae strain YG3-1 yeasts (containing induced heat shock proteins) on the survival of Artemia in stress conditions, was evaluated. For this purpose, heat shock proteins 82 (Hsps 82) o...
